Novo Nordisk's Ozempic: Recent Study Reveals Potential Risk of Rare Vision Loss

Increased Risk of NAION

An in-depth study analyzing years of patient data from Denmark and Norway indicated that diabetes patients using Ozempic were more than twice as likely to suffer from non-arteritic anterior ischemic optic neuropathy (NAION), a rare vision impairment condition, compared to those on a different diabetes medication. The research was published on medRxiv on December 11, highlighting a potential concern about long-term use of the drug.

Understanding NAION and Study Findings

NAION is caused by reduced blood flow damaging the optic nerve, often impacting individuals over 50, and compounded by conditions like diabetes and heart disease. Although the occurrence of NAION identified in the study was low—adding just 1.4 cases per 10,000 years of patient observation—the condition is irreversible and potentially blinding, prompting advice for patients to be informed about these risks.

The study, using data from the University of Southern Denmark, Norwegian Institute of Public Health, and the University of Copenhagen, found a statistical increase in NAION instances among over 60,000 Ozempic users in Denmark and Norway. However, due to fewer cases, the potential risk for those taking its sister drug, Wegovy, particularly prescribed for obesity, remains inconclusive.

Market and Company Responses

Novo Nordisk's confidence in its drug remains unchanged despite the findings, citing that only a few NAION incidences were seen in its trials without tilting against its drugs. Nevertheless, shares in Novo dipped as much as 5.4% in Copenhagen following the publication, though the stock has risen approximately 8% over the year.

Weighing Therapeutic Benefits Against Risks

While the study suggests an Ozempic user's 0.3% to 0.5% risk of developing NAION over a 20-year period, researchers urge balancing the therapeutic benefits of semaglutide, the active ingredient, against this potential vision loss risk. The study's scope, including over 44,000 patients in Denmark and over 16,000 in Norway, marks it as a significant dataset tapping into the post-market safety assessment of Ozempic since its introduction in 2018.
